Glucocorticoids, a group of steroid hormones, are commonly used to treat various inflammatory, autoimmune, allergic, and respiratory conditions. These potent anti-inflammatory and immunosuppressive agents help alleviate symptoms such as swelling, pain, and tissue damage.

In the human body, glucocorticoids work by reducing inflammation and suppressing immune system activity. They inhibit inflammatory cytokines and mediators at the cellular level, blocking pathways like NF-κB, and decreasing enzymes such as phospholipase A2. This reduction in production of pro-inflammatory molecules helps to ease inflammation.

Glucocorticoids also suppress the immune system's function, thereby reducing antibody production and cell-mediated immunity. This is beneficial in autoimmune and allergic diseases, but it does increase the risk of infections. In respiratory conditions, glucocorticoids promote bronchodilation by reducing airway inflammation and remodeling.

Common health conditions treated with glucocorticoids include rheumatoid arthritis and other rheumatic disorders, allergic diseases, autoimmune diseases, respiratory diseases, inflammatory bowel disease flares, prevention of transplant rejection, and adrenal insufficiency treatment.

Examples of glucocorticoid drugs include dexamethasone, prednisone, prednisolone, methylprednisolone, triamcinolone, and hydrocortisone. However, side effects of long-term use can include immunosuppression, which makes a person more vulnerable to infections, suppression of the adrenal glands, high blood pressure, changes in body fat distribution, weight gain or obesity, muscle weakness, stomach ulcers, glaucoma, diabetes, Cushing's syndrome, mood changes, and suppressed growth in children.

Anyone taking glucocorticoids for long periods should consult a doctor for advice on discontinuing use safely, as abrupt discontinuation can cause dangerous side effects. It is also important to contact a doctor if one experiences any of the following while taking glucocorticoids: continued or worsening symptoms of their condition, mental health changes, serious side effects, signs of an infection, or if they wish to discuss safe discontinuation.
